Description

CRISPR is a powerful genomic tool allowing scientists to knock out or add sequences to DNA by use of guide RNAs and Cas nucleases. Guide RNAs target specific sequences ensuring a precise location is affected in the “change” made by CRISPR editing. A Cas nuclease is an enzyme used to snip in or out the desired sequence. A plasmid, PX458, is used to deliver the guide RNA and CAS9 into the cell’s nucleolus where DNA is housed.

A) Blue: DAPY is a fluorescent dye which binds to nucleic acids therefore, identifying the nucleus.

B) Green: GFP gene expressed shows the successful insertion of the plasmid, PX458.

C) Merge of blue and green channels: successful transfection of PX458 suggesting the expression of the gRNA and Cas9.
